Assessment of the Pharmacological and Therapeutic Effect of Holarrhena Antidysenterica in FCA Induced Rheumatoid Arthritis in Wistar Rats

Rheumatoid Arthritis (RA) is a long-term inflammatory circumstance marked via significant joint deterioration and ongoing inflammation. Holarrhena antidysenterica, herb integral to Ayurvedic therapy, shows promising immunomodulatory and anti-inflammatory properties. In this work, Freund's Complete Adjuvant induce rheumatoid arthritis in Wistar rats to evaluate pharmacological & therapeutic potential of Holarrhena antidysenterica bark extract, employing in vitro & in vivo models. The purchase of Holarrhena antidysenterica extract of bark marked beginning investigation, which was then followed by a preliminary phytochemical screening to determine active ingredients. The anti-arthritic properties of substance were assayed using the protein denaturation & HRBC membrane stabilization method in vitro. In vivo, Wistar rats induced with rheumatoid arthritis by FCA were monitored for changes in body weight, joint diameter, paw volume, observatory parameters. Additionally, haematological parameters (RBCs, WBCs, ESR, Hb, platelet count) and X-ray radiography of left hind paw evaluated structural changes. Holarrhena antidysenterica's therapeutic effects were elucidated through assessment of levels of proinflammatory cytokines, CRP, histological changes in ankle joint tissues. Statistical analysis involved ANOVA, Student t-test, and Dunnett's test for evaluation significance to assess significance compared to disease control group. Results show that bark extract of Holarrhena antidysenterica has considerable anti-arthritic feasible in vitro & in vivo, as demonstrated by improvements in biochemical, histological, haematological, radiographic, physical, observational parameters. These results validate traditional usage of Holarrhena antidysenterica and suggest for further studies to see whether it may be used as a rheumatoid arthritis treatment.
